Raman spectroscopy as a tool for monitoring mesoscale continuous-flow organic synthesis: Equipment interface and assessment in four medicinally-relevant reactions

Abstract: SummaryAn apparatus is reported for real-time Raman monitoring of reactions performed using continuous-flow processing. Its capability is assessed by studying four reactions, all involving formation of products bearing α,β-unsaturated carbonyl moieties; synthesis of 3-acetylcoumarin, Knoevenagel and Claisen–Schmidt condensations, and a Biginelli reaction. In each case it is possible to monitor the reactions and also in one case, by means of a calibration curve, determine product conversion from Raman spectral … Show more

“…Several in-line monitoring tools, for example, ReactIR [58][59][60][61][62] and flow based NMR [63][64][65][66] which allow for substrate specific, nonconsuming analysis, have also been developed. Other in-line techniques employing simple UV [67][68][69][70], MS [71][72][73], and Raman [74][75][76][77] analysis have also been successfully applied enabling efficient optimisation and generation of new reaction chemistry.…”

“…The use of Raman spectroscopy has recently been reported for the continuous monitoring of mesofluidic scale flow syntheses 41. An off‐the‐shelf spectroscopic flow cell was placed into an assembly that enabled the position of the Raman light source to be adjusted to optimize the intensity of the gathered signal.…”

“…Über Raman‐Spektroskopie zur kontinuierlichen Verfolgung von Strömungssynthesen im mesofluidischen Maßstab wurde kürzlich berichtet 41. Eine handelsübliche spektroskopische Durchflusszelle wurde in einer Anordnung platziert, die es gestattete, die Position der Raman‐Lichtquelle so zu ändern, dass sich die Intensität des erhaltenen Signals optimieren ließ.…”
