Rap1 mediates sustained MAP kinase activation induced by nerve growth factor

Abstract: Activation of mitogen-activated protein (MAP) kinase (also known as extracellular-signal-regulated kinase, or ERK) by growth factors can trigger either cell growth or differentiation. The intracellular signals that couple growth factors to MAP kinase may determine the different effects of growth factors: for example, transient activation of MAP kinase by epidermal growth factor stimulates proliferation of PC12 cells, whereas they differentiate in response to nerve growth factor, which acts partly by inducing a… Show more

“…We find that G αi -Rap1-Src-STAT-3 pathway plays a major role in neurite outgrowth induced upon activation of a number of these receptors [102][103][104]. This is consistent with studies in PC12 cells where Rap1 and STAT-3 have been implicated [105,106]. In addition, muscarinic receptor induced neurogenesis of neural stem/progenitor cells involves CREB signaling which is mediated by Src phosphorylation [107].…”

“…In contrast, factors such as epidermal growth factor which elicit transient ERK activity are mitogenic [52]. Both epidermal growth factor and nerve growth factor induce transient ERK activation via Ras and Raf-1, but the latter can procure the sustained activation of the ERK pathway via the activation of BRaf by Rap1 [53]. This neat example of achieving a specific biological response is based on the switching of allegiances from a Ras-Raf-1 to a Rap1-B-Raf signalling complex.…”

“…Introduction cAMP inhibits cellular proliferation by interfering with the mitogen-activated protein (MAP) kinase pathway at the cytosol (Wu et al, 1993;Cook and McCormick, 1993;Burgering et al, 1993;Chen and Iyengar, 1994;Hordijk et al, 1994;York et al, 1998). The nuclear events by which cAMP exerts such growth-inhibitory e ect remain largely undetermined (Xing et al, 1996;Nakajima et al, 1996).…”
