Rapid detection of praziquantel using monoclonal antibody-based ic-ELISA and immunochromatographic strips

Abstract: Praziquantel (PZQ) is a broad-spectrum antiparasitic drug used in mammals and fish. In this study, we developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for the detection of PZQ residues. The hapten PZQ-HS produced by introducing an amino group at a benzene ring synthesized the immunogen and the coating antigen with carrier proteins. Highly sensitive monoclonal antibodies (mAbs) were prepared following mouse immunization and cell fusion. Under optimum conditions, the half maximal in… Show more

“…Unlike ELISAs, all reagents are sprayed onto the test strip with the LFIAs (Guo et al, 2019;Liu et al, 2019;Song et al, 2019;Xu et al, 2019). Rapid detection of PZQ using ELISAs and LFIAs in mackerel samples has been developed recently with a 50 ng/mL limit of detection (LOD) (Shen et al, 2019). In this study however, we aim to develop a more sensitive assay to detect PZQ by optimizing the strip test for use in the Japanese sea bass (Lateolabrax japonicus).…”

“…Generally, immunochemical detection methods have proven to be simple, cost-effective and rapid (Pang et al, 2019;Song et al, 2018). The enzyme-linked immunosorbent assay (ELISA) is a rapid detection method that utilizes the specific reaction of an antigen with an antibody (Shen et al, 2019;Ye, Wu, Xu, Zheng, & Kuang, 2018). In addition, immunoassay kits that now widely utilized in food analysis are portable and can be used for routine in a processing facility or retail outlet.…”

“…In general, chromatography and mass spectrometry such as gas chromatography, high performance liquid chromatography, and liquid chromatography-tandem mass spectrometry (Chen et al, 2016;Huang et al, 2019;Ling-Li et al, 2016;Rafique et al, 2016;Reinholds et al, 2016) have excellent sensitivity and reliability for TZP detection. However, these methods are expensive, time consuming, and require experienced technicians (Chao et al, 2020;Chen et al, 2019;Shen et al, 2019).…”
