Rapid Procedure for Estimating Cyclic Hydroxamate (DIMBOA) Concentration in Maize1

Abstract: A rapid procedure was developed for estimating the concentration of a cyclic hydroxamic acid 2,4‐dihydroxy‐ 7‐methoxy‐l,4‐benzoxazin‐3‐one (DIMBOA) for potential use in a maize (Zea mays L.) breeding program. Samples (0.2 to 0.5 g stem tissue) were collected from seedlings at 36 cm to 41 cm extended leaf height. The rapid procedure, essentially a simplification of the colorimetric procedure of Hamilton, was significantly correlated with the latter (r = +0.82) based on the analysis of 13 inbred lines of maize.

“…However, these have concentrated on seedlings. Comparisons between analytical techniques should be made for flag leaves when the possibility of erroneous results using the non-specific ferric chloride method is increased due to the higher concentrations of phenols in mature plants (Long, Dunn & Routley, 1974;Leszczynski et al, 1989).…”

“…A simple colorimetric test (Long et al. ) was used to confirm the benzoxazinoid‐synthesis phenotype of each line. Ten plants from each line were germinated in moist vermiculite and allowed to grow for 7 days.…”

“…Plants of each line were grown in two separate greenhouses and self-pollinated to provide sufficient seed for experiments. A simple colorimetric test (Long et al 1974) was used to confirm the benzoxazinoid-synthesis phenotype of each line. Ten plants from each line were germinated in moist vermiculite and allowed to grow for 7 days.…”
