Rat ovulation, implantation and decidualization are severely compromised by COX-2 inhibitors

Abstract: Although Cyclooxygenase-2 (COX-2) is essential for mouse ovulation, fertilization, implantation and decidualization, the regulation and function of COX-2 in rat reproduction are still unknown. This study was designed to examine the action of COX-2 in rat ovulation, implantation and decidualization by using two specific inhibitors of COX-2 (nimesulide and niflumic acid). Compared to control, either nimesulide or niflumic acid significantly inhibited the ovulation in the superovulated rats. Although nimesulide h… Show more

“…Further study found that Ahr could induce transcription of Cox2 in breast cancer MCF7 cells (Degner et al 2007). Cox2 is an inducible rate-limiting enzyme in the biosynthesis of prostaglandins and implicated in mouse decidualization (Lim et al 1997, Diao et al 2007, Zhang et al 2013. Inhibition or deficiency of Cox2 could result in defective decidualization (Lim et al 1997, Diao et al 2007.…”

“…Numerous data declared that endometrial vascular permeability and angiogenesis were profoundly influenced by prostaglandins (PGs) which were the product of a cascade of enzymes such as Cox and PG synthases. 23,33 Cox-2 was an inducible rate-limiting enzyme in the biosynthesis of PGs and could convert arachidonic acid into PGH 2 , a common precursor of all PGs. 1,33 The targeted disruption of Cox-2 in mice led to the failure of implantation.…”

“…Because endometrial vascular permeability has been considered as a marker for embryo implantation, 23 intrauterine blocking treatments were used to examine whether Egr1 could regulate the vascular permeability of mouse implantation sites. After the intrauterine injection of Egr1-specific siRNA, vascular permeability of the implantation sites on day 5 of pregnancy was significantly declined compared to that of scrambled siRNA group (Fig.…”

“…As well, several nonsteroidal anti-inflammatory drugs (NSAIDs) and selective COX-2 inhibitors were implicated in the inhibition of endometrial vascular permeability and implantation in a variety of species (Diao et al, 2007;Sookvanichsilp & Pulbutr, 2002).…”
