Rationale for transdermal drug administration in Alzheimer disease

Abstract: Transdermal patches are used for the treatment of various diseases including neurologic and psychiatric disorders such as Parkinson disease (PD), major depression, and attention deficit hyperactivity disorder. They are believed to offer many advantages over conventional oral therapies. By providing smoother, continuous drug delivery and steadier plasma levels, patches may reduce the incidence of side effects, thus making optimal therapeutic doses easier to attain and potentially improving treatment efficacy an… Show more

“…Even in unconscious or nauseated patients, it is possible to terminate drug input simply by washing or peeling (21,22). A wide range of transcutaneous products have been approved, providing acute and chronic treatment modalities for menopause and postmenopausal osteoporosis, contraception, angina, pain, smoking cessation, motion sickness, dementia, Alzheimer's disease, Parkinson's disease, attention deficit disorder and asthma (23,24). These attest to the success of delivery via the skin and have been very effective adjuncts to oral therapies.…”

“…Since the reported antioxidant and anti-inflammation activity of ZC oil is associated with AD, investigation of the anti-cholinesterase activity of ZC oil would be an interesting aspect to confirm the possibility of using ZC oil in the treatment of AD. Moreover, incorporation of ZC oil in a novel topical formulation will also be fascinating because transdermal administration is the ideal therapeutic approach for chronic neurological disorders in elderly people since it provides sustained therapeutic plasma levels of drugs, is simple to use, and may reduce systemic adverse effects (23). ZC oil is a complex mixture of volatile components and its constituents can degrade during storage and lead to changes in biological activities.…”
