Reactivation of endogenous retroviral elements via treatment with DNMT- and HDAC-inhibitors

Abstract: Inhibitors of DNA methyltransferases (DNMTis) or histone deacetylases (HDACis) are epigenetic drugs which are investigated since decades. Several have been approved and are applied in the treatment of hematopoietic and lymphatic malignancies, although their mode of action has not been fully understood. Two recent findings improved mechanistic insights: i) activation of human endogenous retroviral elements (HERVs) with concomitant synthesis of double-stranded RNAs (dsRNAs), and ii) massive activation of promote… Show more

“…These epigenetic modifications may explain the polarization profiles such as tolerance or trained immunity, observed after various stimulations of innate cells (Saeed et al 2014). We hypothesized and confirmed in vivo that other elements than genes, especially HERVs which are known to be tightly controlled by epigenetic modifications (Daskalakis et al 2018), might be modulated in acute inflammatory situations. This has also been demonstrated in other pathophysiological contexts such as cancer (J.…”

“…HDACi can modify the acetylation status of histones, resulting in the induction of cell cycle arrest, apoptosis or differentiation ( Eckschlager et al, 2017 ). HDACi potentially re-activate HERVs, however, the beneficial or detrimental effects of epigenetic drugs on HERV modulation are currently discussed ( Chiappinelli et al, 2015 ; Hurst et al, 2016 ; Daskalakis et al, 2018 ; White et al, 2018 ).…”
