Intestinal bacteria release various neuroactive compounds directly or indirectly regulating brain function to modulate host health and behavior through the gut-brain axis. Probiotics have been used as dietary supplements to target gut microbiota (microbiome) for prevention or therapeutic treatment of various diseases including mental disorders. In our study, chickens were used as an animal model to assess, if dietary supplementation of probiotic, Bacillus subtilis, reduces aggressive behaviors following social challenge. Chickens of an aggressive line were housed in single-hen cages. At 24 weeks of age, the hens were paired with similar body weight to identify the dominance rank (day 0). The subordinate and dominant of each pair were fed a regular layer diet or the diet mixed with 250 ppm probiotics for 2 weeks, then the second behavior test was performed between the same pair (day 14). The display of aggressive behaviors in the regular diet-fed chickens was not affected between the levels at day 0 and day 14, while the frequency of threat and aggressive pecking were reduced in the probiotic-fed chickens compared to the levels at day 0. These results suggest dietary probiotic, Bacillus subtilis, could be a suitable strategy for increasing hosts' mental health.

Dysbiosis is a condition that can cause various clinical disorders, from gastrointestinal problems to allergies or even cancer. Resetting the microbiota using antibiotics and/or probiotics could be a possible therapy for many diseases. The aim of this study was to evaluate the effects of three treatment regimens in patients with irritable bowel syndrome (IBS). The regimens were short-term rifaximin treatment (10 days) followed by either a nutraceutical agent (G1) or a low- Fermentable, Oligo-, Di-, Monosaccharide and Polyol (FODMAP) diet (24 days) (G3) or treatment with MegaSporeBiotic a mixture of spores of five Bacillus spp. for medium-term (34 days) (G2). Ninety patients with IBS without constipation were enrolled and divided into three groups (G1, G2, G3). Patients in G1 and G3 were evaluated over four visits (baseline/first day (V1), 10 days (V2), 34 days (V3), 60 days (V4)), and, those in G2 over three visits (V1, V3, V4). Severity score, quality of life, and parameters from the rectal volume sensation test were determined. The results demonstrated that patients treated with MegaSporeBiotic, compared with those treated with rifaximin followed by nutraceutical or low-FODMAP diet, had similar severity scores and rectal volume sensation test results for all parameters tested and statistically significant improvement in measurements of quality of life.

This study was conducted to investigate the effects of dietary supplementation with a mixture of Bacillus on the intestinal health of weaned piglets. We randomly assigned 120 piglets to three groups; a control group (basal diet), a probiotics group (supplemented with 4 × 109 CFU/g Bacillus licheniformis-B. subtilis mixture; BLS mix), and an antibiotics group (supplemented with 0.04 kg/t virginiamycin, 0.2 kg/t colistin, and 3000 mg/kg zinc oxide). All groups had five replicates with eight piglets per replicate. On days 7, 21, and 42 of the trial, intestine and digesta samples were collected to determine the intestinal morphology, gut microbiota and metabolites, and the expression of genes related to gut health. The results showed that the BLS mix decreased the jejunum crypt depth, increased the ileum villus height, and increased the jejunum and ileum villus height to crypt depth ratio. The BLS mix also increased the expression levels of E-cadherin and Occludin in the colon and pro-inflammatory cytokines and TLR4 in ileum and colon. The BLS mix also increased Simpson’s diversity index in the gut microbiota and the relative abundances of o_Bacteroidetes and f_Ruminococcaceae, but it decreased the relative abundances of Blautia, and Clostridium. Collectively, these findings suggested that dietary BLS mix supplementation efficaciously promotes intestinal health through the modulation of gut microbiota in weaned piglets.
