Recent Advances on Small-Molecule Survivin Inhibitors

Abstract: Survivin, a member of the inhibitor of apoptosis proteins family, is highly expressed in most human neoplasms, but its expression is very low or undetectable in terminally differentiated normal tissues. Survivin has been shown to inhibit cancer cell apoptosis and promote cell proliferation. The overexpression of survivin closely correlates with tumor progression and drug resistance. Because of its key role in tumor formation and maintenance, survivin is considered as an ideal target for anticancer treatment. H… Show more

“…As a key pro-survival factor, survivin is overexpressed in most solid tumors [ 7 ]; therefore it has been actively pursued as a promising target to develop effective cancer therapy [ 34 36 ]. Various strategies are being explored, including vaccination strategies to generate an antigen-specific immune response against survivin-bearing tumor cells; the development of antisense oligonucleotides, ribozymes, or siRNA molecules targeting survivin; and small-molecule inhibitors of survivin function [ 37 ].…”

“…Several small molecule inhibitors targeting Survivin have also been evaluated in various in vitro and in vivo studies. Despite the challenges associated with small molecule drug development, these inhibitors are important for understanding Survivin biology and anti-cancer drug development [ 134 , 135 ]. YM-155 is a novel small molecule which suppresses transactivation of Survivin through direct binding to its promoter [ 136 ].…”

“…Therefore, it is considered as a cancer-specific biomarker. Hence, targeting survivin embodies an attractive strategy for the development of more selective anticancer therapeutics [ 8 , 9 , 10 , 11 , 12 , 13 , 14 , 15 , 16 ].…”
