“…There were such observations reported before Garnier's thesis (see Altmann (3), De Groot, La Gasse, and Sebruyns (17), Henry (27), Krabbe (31), and Wilson (52) for some of the older literature) and many additional studies have been reported during the 60 year interval between the work of Gamier and Hsu and Lou and Wendt. Various aspects of the nudeocytoplasmic relationship have been covered in recent reviews (Baud (7), Bloch (10), Briggs and King (14), Gates (22), Hertl (28), Kurnick (32), Mazia (36), Stich (44), and Vincent (47)), and no attempt will be made here to review the subject. The admirable report of Hertl (28) carefully compares many of the opinions about the structure and function of the nucleolus (nucleolar apparatus) and discusses the transfer of nuclear and nucleolar material into the cytoplasm.…”

“…At least 3 anatomic mechanisms of such extrusions have been described. (a) A nuclear vesicle ("bleb" or "pocket") is said to form at the nuclear membrane and the "bleb" then ruptures or is dispersed in some fashion into the cytoplasm (7,30,31,51). (b) The nuclear membrane is described as opening temporarily ("Schleusenmechanismus" of Berg) and such openings are associated with transfer of nuclear material into the cytoplasm (3)(4)(5)8).…”

“…However, the fact that this does not hold true for other material may be the reason why, in cases where the membrane is seen only in fixed condition, doubt has arisen as to the existence of an in vivo nuclear membrane. With this in mind Baud (1953) appears to be justified in his conclusion that the nuclear membrane is visible in some cells in the living condition, while in other cells indirect methods, such as phase contrast microscopy and microdissection, can be employed to show its presence. In spite of the fact that it cannot be demonstrated under all conditions in all material, the existence of the nuclear membrane has generally been accepted as representing an in vivo structure separating the nuclear con tents from the cytoplasm.…”

“…Another view long since abandoned, which was held by early microscopists (see Wilson 1937), claimed that the nuclear membrane was simply a denser portion of the protoplasm, with the nucleus being conceived only as a localized area within the common structural framework. Baud (1953) has discussed this problem at length and has shown that the existance of the nuclear membrane can be established by the fact that it is ob servable under in vivo conditions employing phase contrast optics (Zollinger 1948). Such evidence is strengthened by the findings obtained by the use of in direct procedures, the most convincing of which is microdissection (Chambers 1924).…”
