Recombinant Allergens for Skin Testing

Abstract: Skin testing is a basic diagnostic procedure widely used to explore immediate-type reactions to allergen preparations in vivo. Despite their reliability, if standardized extracts are used, skin tests suffer from limited reproducibility due to difficulties in preparing consistently standardized extracts from natural raw material. Starting from allergen-encoding cDNAs, large amounts of highly pure allergens with a high batch-to-batch consistency satisfying the quality requirements of medicinal products manufactu… Show more

“…Their sensitivity, however, appears to be lower when compared to natural allergen extracts. 89,96 Which allergens to test is a common doubt in daily practice. A recent survey performed in the United States showed that most allergists do not rely on history when choosing which allergens to use to perform skin testing.…”

“…However, only a few comparative SPT studies with purified food allergens have been conducted, [28], [72], [73] and [74] which is mainly due to the high demands of approval in clinical trials as requested by the institutional review boards and regulatory authorities to ensure patient safety (test allergens have to be licensed as biotechnologic products). These few studies verified a superior diagnostic potential of recombinant allergens.…”

“…Hence, skin tests with recombinant molecules need to include panels of recombinant allergens covering all immunodominant structures present in a given food. [72] and [74] …”
