Recombinant HLA-DP2 Binds Beryllium and Tolerizes Beryllium-Specific Pathogenic CD4+ T Cells

Abstract: Chronic beryllium disease is a lung disorder caused by beryllium exposure in the workplace and is characterized by granulomatous inflammation and the accumulation of beryllium-specific, HLA-DP2-restricted CD4+ T lymphocytes in the lung that proliferate and secrete Th1-type cytokines. To characterize the interaction among HLA-DP2, beryllium, and CD4+ T cells, we constructed rHLA-DP2 and rHLA-DP4 molecules consisting of the α-1 and β-1 domains of the HLA-DP molecules genetically linked into single polypeptide ch… Show more

“…An anti-LFA-1 antibody was shown to inhibit cell proliferation and cytokine release in a CD4 + T cell line derived from a CBD patient stimulated with antigen presenting cells and BeSO 4 . 38 Stimulated CD4 + T cell lines also exhibited a marked increase in cell surface expression of I-CAM1 within 24 h following stimulation. These studies suggest that LFA-1 and I-CAM1 interactions play an important role in the cell-cell adhesion processes involved in CBD disease progression and granuloma formation.…”

“…General methods for the design, cloning, and expression of RTLs have been described previously (Huan et al, 2004;Burrows et al, 1999;Fontenot et al, 2006). In brief, a series of murine MHC class II I-A b -derived single-chain ␤1/␣1 recombinant RTLs, termed rIA b (RTL550), were constructed by sequential site-directed mutagenesis of rIA q RTLs.…”

“…The rIA b /MOG insert was ligated into pET21d(ϩ) vector and transformed into Nova blue Escherichia coli host (Novagen, Madison, WI) for positive colony selection and sequence verification, and rIA b / MOG plasmid constructs were then transformed into the E. coli strain BL21(DE3) expression host (Novagen). Protein purification was as described previously (Huan et al, 2004;Burrows et al, 1999;Fontenot et al, 2006) with a 30 -40 mg yield of purified protein per liter of bacterial cell culture.…”

“…When Be-reactive lung T cells are tested for an in vitro response to Be, the DP alleles implicated in disease susceptibility are the same as those shown to present Be to T cells (18,19). In addition, soluble HLA-DP molecules expressing βGlu69, but not HLA-DP molecules with a lysine at that position, can bind beryllium in vitro with high affinity (20,21). Thus, these findings strongly suggest that the ability to bind and present Be is the reason for the association of certain HLA-DP alleles and disease susceptibility.…”
