Recombinant human bone morphogenetic protein‐2 is superior to demineralized bone matrix in repairing craniotomy defects in rats

Abstract: The purpose of this study was to measure bone-regenerative effects of recombinant human bone morphogenetic protein-2 (rhBMP-2) in rat calvarial critical-size defects (CSDs). CSDs (8 mm in diameter) were treated with either 1) 2.2 micrograms rhBMP-2 combined with insoluble collagenous bone matrix (ICBM), 2) 6.5 micrograms rhBMP-2 plus ICBM, 3) ICBM alone, or 4) demineralized bone matrix (DBM), for 7, 14, or 21 days. Multiple linear regression showed that rhBMP-2 had a significant time- and dose-dependent effect… Show more

“…BMP-2 is superior to DBM, also in rats (Marden et al 1994). The BMP-2 and collagen device used in this study has proven efficient in critical-sized long bone and mandibular defects in rodents, dogs and monkeys (manuscripts in preparation).…”

“…These studies comprise the grafting of a segmental bone defect that does not heal spontaneously during the lifetime of the animal; such a defect is also called a critical-size defect. Complete healing of critical-size defects in rat calvariae was observed after grafting fleeces of bovine bone derived collagen that had been soaked in a rhBMP-2 solution (95). Calcifying cartilage and remineralization of the collagen carrier were observed after 1 week, which was followed by rapid bone formation (95).…”

“…Devido à potente ação osteoindutora observada nestes estudos, torna-se evidente o potencial dessas proteínas para uma grande variedade de aplicações clínicas como cirurgias reconstrutivas, defeitos ósseos provocados por doenças, perdas ósseas devido a alterações fisiológicas, dentre outras situações que requerem o reparo ósseo (BOYNE, 1995;MARDEN et al, 1994).…”
