Recombinant Protein-Based Assays for Detection of Antibodies to Severe Acute Respiratory Syndrome Coronavirus Spike and Nucleocapsid Proteins

Abstract: Recombinant severe acute respiratory syndrome (SARS) nucleocapsid and spike protein-based immunoglobulin G immunoassays were developed and evaluated. Our assays demonstrated high sensitivity and specificity to the SARS coronavirus in sera collected from patients as late as 2 years postonset of symptoms. These assays will be useful not only for routine SARS coronavirus diagnostics but also for epidemiological and antibody kinetic studies. The 2003 outbreak of severe acute respiratory syndrome (SARS) lasted fewer… Show more

“…His-tagged recombinant protein was expressed in Escherichia coli and purified by metal affinity chromatography. Recombinant HKU5.2N protein indirect ELISA was developed using a modified version of the severe acute respiratory syndrome (SARS) CoV N ELISA described by Haynes et al [5]. Sera were considered positive when the optical density (OD) values were at or above the 0.43 cutoff value (mean absorbance at 405 nm of sera from US blood donors plus 3 standard deviations).…”

“…The virus is a (+) sense single-stranded RNA virus with a genome containing 29,727 nucleotides (2,3) which consist of four structural proteins: spike (S), envelope (E), membrane (M), and nucleocapsid (N) (4). Similar to other coronaviruses, the S and N proteins are highly expressed during viral infection and are a useful target for immune detection (5,6). The SARS CoV N protein is of particular interest because it can be a potential target for vaccine development (7) as well as a diagnostic marker for SARS CoV infection (8,9).…”
