Redox regulation of ascorbate and glutathione by a chloroplastic dehydroascorbate reductase is required for high-light stress tolerance in Arabidopsis

Abstract: Chloroplasts are a significant site for reactive oxygen species production under illumination and, thus, possess a well-organized antioxidant system involving ascorbate. Ascorbate recycling occurs in different manners in this system, including a dehydroascorbate reductase (DHAR) reaction. We herein investigated the physiological significance of DHAR3 in photo-oxidative stress tolerance in Arabidopsis. GFP-fused DHAR3 protein was targeted to chloroplasts in Arabidopsis leaves. A DHAR3 knockout mutant exhibited … Show more

“…A minor contribution to the cytosolic activity would come from DHAR2. This is consistent with two recent reports on single dhar mutants (Noshi et al, 2016(Noshi et al, , 2017 but in contrast to an earlier analysis of dhar2 (Yoshida et al, 2006).…”

“…We cannot discount a role for DHARs in maintaining ascorbate in other conditions. For example, changes in ascorbate redox state were reported in dhar1 and dhar2 single mutants subjected to high light (Noshi et al, 2016(Noshi et al, , 2017. However, these effects were relatively slight.…”

“…However, the ROS can be scavenged by plant antioxidant defense system consisting of antioxidant compounds and enzymes (Apel and Hirt, 2004; Suzuki et al, 2012). The ascorbate (ASA)–glutathione (GSH) cycle is one of the main antioxidant systems in plants to keep ROS under control being, which involves a lot of critical antioxidant enzymes in a series of cyclic reactions to detoxify H 2 O 2 (de Sousa et al, 2016; Noshi et al, 2016). One of the antioxidant enzymes is peroxidase (POD), which can detoxify H 2 O 2 by oxidizing ascorbate.…”

“…() that mutants deficient in pyrroline‐5‐carboxylate synthase‐1, having lower proline content, showed reduced activity of the ascorbate‐glutathione cycle, which is an important ROS scavenging pathway (Foyer and Noctor, ; Noshi et al. , ). The higher proline content in L er together with its faster association with the chloroplasts constitutes a clear advantage of the wildtype and may, at least in part, explain the light sensitive phenotype of gin2‐1 .…”
