Redox-Sensitive Polymer/SPIO Nanocomplexes for Efficient Magnetofection and MR Imaging of Human Cancer Cells

Abstract: Magnetofection has received increasing attention for its great potential on gene therapy. To promote its clinical therapeutic applications, development of safe and effective magnetic nanocarriers is in high demand. Herein, we present a redox-sensitive polymer/metal nanocomplex system (PSPIO) for efficient magnetofection and magnet resonance imaging (MRI) on cancer cells. PSPIO was prepared by modifying SPIO with redox-sensitive polyethylenimine (SSPEI) via a ligand exchange process. PSPIO could efficiently con… Show more

“…Dual/multi‐responsive delivery systems have the ability to simultaneously overcome various barriers during targeted delivery, including blood clearance, cellular uptake and intracellular release of the cargo. Among the designs of most multi‐responsive systems, the most commonly studied comprise a combination two different stimuli, such as pH/redox, pH/ROS, pH/ATP, pH/enzyme, pH/light, light/redox and redox/magnetic field . Those systems have some unique characteristics, including high cell uptake, low cytotoxicity and specific intracellular gene release, etc.…”

“…Several attempts, also including NPs, have been made to distribute anti-survivin siRNA in order to silence the BIRC5 gene [445,446,447]. For NP-based GTH applications for PCa, there are also quite few publications the last decade [141,448,449,450,451,452]. Guo et al investigated, for example, gene silencing using siRNA-based AuNPs for LNCaP cells, overexpressing PSMA.…”

“…The in vivo transfection efficiency was greatly increased under the influence of external magnetic field. This polymeric SPIONs demonstrated significant transgene expression on human prostate cancer cell lines and mouse glioma . Prijic et al used surface modified magnetic nanoparticles against murine mammary cancer.…”
