Reflections on the clonal-selection theory

Abstract: How do we account for the immune system's ability to produce antibodies in response to new antigens? It has been 50 years since F. Macfarlane Burnet published his answer to this question: the clonal-selection theory of antibody diversity. The idea that specificity for diverse antigens exists before these antigens are encountered was a radical notion at the time, but one that became widely accepted. In this article, Nature Reviews Immunology asks six key scientists for their thoughts and opinions on the clonal-… Show more

“…Another important concept in early adaptive immunity is that of clonal expansion [18], and shedding at the cell surface influences lymphocyte proliferation by modulating ligand receptor interactions. LAG3 (Lymphocyte Activation Gene 3) inhibits T-cell proliferation, and its cleavage by ADAM10 and ADAM17 alters clonal expansion ( Figure 2a).…”

“…In the case of B lymphocytes, antigen receptors can be further diversified in an antigen-dependent manner by somatic hypermutation of clonally expanded lymphocytes. 2,3 Thus, the basic unit of the B cell repertoire is the idiotype, which refers to a unique antigen receptor structure (H and L chain pair) with unique epitope specificity. One or more idiotypes derived from the same VDJ recombination event and H C L pairing represents a clonotype.…”

“…В основе обсуждаемой парадигмы лежит теория клональной селекции, предложенная Бернетом в се-редине прошлого века, активно дискутировавшаяся и модифицированная современными учеными [60,61]. Одним из важнейших постулатов этой теории остается утверждение о том, что В-клеточный от-вет, приводящий к продукции антител, вызывается не любым чужеродным антигеном, а лишь теми, в контексте молекулярной структуры которых ВкР антителопродуцента могут отличить «свое» от «не своего».…”
