Regulation of cell proliferation, inflammatory cytokine production and calcium mobilization in primary human T lymphocytes by emodin from Polygonum hypoleucum Ohwi

Abstract: We hypothesize that the inhibitory mechanisms of emodin on activated T cells proliferation are related to the impairment of cytokine production, IL-2 mRNA level and [Ca2+]i in the cells.

“…Emodin isolated from a great deal of herbs is an effective constituent with many effects [3][4][5] . It was reported that emodin could inhibit the expression of α-smooth muscle actin (α-SMA) and reduce the transformation and synthesis of TGF-β1 [6] .…”

“…Emodin (1, 3, 8-trihydroxy-6-methylanthraquinone) isolated from a g reat deal of herbs is an effective constituent with many effects. Lots of pharmaceutical studies have demonstrated that emodin has many biological effects [3][4][5] , such as anticancer, antimicrobial, and antiinflammatory effects, etc. Moreover, several studies [6,7] have revealed that emodin is efficacious in the management of acute pancreatitis and hepato-cirrhosis, but studies on chronic pancreatitis are rare.…”

“…Rheum undulatum L. (Polygonaceae, RU), a well known Chinese medicine, is a perennial herb that is distributed and cultivated mainly in South Korea (9). The rhizome of the species is one of the important herbal medicines that are used widely as laxatives, anti-inflammatories and anti-blood stagnation agents in East Asia (10,11). Recently, it was found that RU and 3 compounds isolated from it (desoxyrhapontigenin, emodine, and chrysophanol) showed significant antidiabetic activity based on the oral glucose tolerance test (9).…”

“…Emodin is the main active component of Da Huang. Previous studies demonstrated that emodin affects bacteriostasis, catharsis, relieves Oddi sphincter spasm, inhibits abnormal metabolism of vasoactive substances (e.g., eicosenoic acid), improves the microcirculation and antagonizes coagulation and thrombus formation (25,29,30). The present study demonstrated that emodin significantly increases MФ ICAM-3 protein expression in rats with SAP/SIRS (P<0.05), thereby promoting the interaction between ICAM-3 and CD14, enhancing identification and phagocytotic capacity of pMФs and relieving inflammatory reactions.…”
