Relationship Between the Gastrointestinal Side Effects of an Anti-Hypertensive Medication and Changes in the Serum Lipid Metabolome

Abstract: An earlier study using a rat model system indicated that the active ingredients contained in the anti-hypertensive medication amlodipine (AMD) appeared to induce various bowel problems, including constipation and inflammation. A probiotic blend was found to alleviate intestinal complications caused by the medicine. To gain more extensive insight into the beneficial effects of the probiotic blend, we investigated the changes in metabolite levels using a non-targeted metabolic approach with ultra-performance liq… Show more

“…occurred due to the induced microbiota compositional change in amlodipine versus amlodipine and probiotic treated mice. 80 Lipid metabolites in amlodipine treated mice had increased levels of phosphatidylcholines, triglycerides with large numbers of double bonds, and cholesterols. The amlodipine treated group also had decreased levels of triglycerides with small numbers of double bonds.…”

“…The authors believed this to be evident that amlodipine shifted the composition and balance of bacterial produced metabolites that ultimately contribute to the GI side effects. 80 Similarly, the calcium channel blockers nifedipine and nimodipine have been investigated for microbial attenuation. Nifedipine and nimodipine have similar mechanisms of action in the treatment of hypertension, and thus, evidence for one drug's relationship with the gut microbiome is potential evidence for the other.…”

“…Recent studies have discovered that metabolites in peripheral blood are associated with the development of liver disease [6], arthritis [7], chronic kidney disease [8], and Alzheimer's disease [9]. In patients with HTN, altered serum metabolic signatures may involve metabolism of fatty acid, glycerophospholipid, alanine, aspartate, and glutamate [10,11]. In addition, significantly different metabolites in HTN have been identified in a pilot study and a randomized clinical trial [12,13].…”

“…Soy protein intake whether in the morning or evening led to a greater microbiota diversity and a decrease in cecal pH resulting from SCFA production compared with casein intake [16]. Finally, Ahn et al [17] investigated the metabolomics changes in rats that received amlodipine. Serum levels of phosphatidylcholine, lysophosphatidylcholine, sphingomyelin, triglycerides with large numbers of double bonds, cholesterol, sterol derivatives, and cholesterol esters were increased.…”

“…Serum levels of phosphatidylcholine, lysophosphatidylcholine, sphingomyelin, triglycerides with large numbers of double bonds, cholesterol, sterol derivatives, and cholesterol esters were increased. Amlodipine-induced compositional changes in the gut microbiota are a causal factor in inflammation [17].…”
