Relationships between Hepatitis C Virus Replication and CXCL-8 Production In Vitro

Abstract: The chemokine CXCL-8 (interleukin-8) is induced by many viruses, including hepatitis C virus (HCV).In the current study, we examined CXCL-8 levels in the context of acute and chronic HCV replication in vitro. Two different small interfering RNAs were used to silence CXCL-8 mRNA and protein expression in Huh7 and BB7 replicon cells. HCV RNA synthesis in BB7 cells was inhibited by CXCL-8 knockdown. Furthermore, antibody neutralization of endogenous CXCL-8 activity inhibited HCV replication, while addition of rec… Show more

“…The rates of annual decline in FEV1 and diffusing capacity for carbon monoxide in COPD patients were significantly higher in HCV-positive patients than in HCV-negative patients (25). These declines were also higher in HCV-positive asthmatic patients than in HCV-negative and interferon responding asthmatic patients (26 (27,28), and that the IL-8 levels are associated with HCV replication (29). The enhanced production of IL-8 is induced via transcriptional ac-T a b l e 3 .…”

“…The most plausible molecule is interleukin (IL)-8, which is one of the key molecules in the pathogenesis of COPD. Hepatitis C virus infection induces IL-8 production in human endothelial cells (18,19), and the IL-8 levels are associated with HCV replication (20). The enhanced production of IL-8 is induced via transcriptional activation and mRNA stabilization (18).…”

“…Huh7.5.1 cells were grown in Huh7 medium as described (9). JFH-1 viral stock preparation, cell infection, and titration was performed as described (33)(34)(35). For antiviral assays, cells were in infected at a multiplicity of infection of 0.05 and protein and RNA harvested 72 h postinfection.…”
