Replication Study to Confirm the Role of CYP2D6 Polymorphism rs1080985 on Donepezil Efficacy in Alzheimer's Disease Patients

Abstract: Alzheimer's disease (AD) is a neurodegenerative disorder often treated with donepezil, an acetylcholinesterase inhibitor. Response to donepezil is variable, probably based on patients' genetic background in donepezil metabolizing enzymes, including cytochrome 2D6 (CYP2D6). We evaluated the association between clinical response to donepezil and a common variant (rs1080985) of CYP2D6, previously reported to be associated with poor response to the drug. In a sample of 415 AD cases, we found evidence of associatio… Show more

“…In one of these studies, a marginal significance for frequency of the APOE ε4 and rs1080985 G allele was found, but an independent effect of APOE ε4 on response to donepezil was not found 8. In our study, APOE ε4 was not significantly associated with the therapeutic response to donepezil.…”

“…Two studies that analyzed the interaction between APOE status and rs1080985 did not find a direct interaction between APOE and CYP2D6 polymorphisms 7,8. “…CYP2D6 variants affect donepezil effi cacy and safety in AD [3,13,17,18,22,26,53,57]. The common variant rs1080985 of CYP2D6 is associated with poor response to donepezil [58,59]. A higher frequency of mutated CYP2D6 allele *2A was found in responder than in nonresponder patients (75.38 vs 43.48%) [60].…”
