Restriction fragment length polymorphisms associated with growth hormone and prolactin genes in Holstein bulls: evidence for a novel growth hormone allele

Abstract: Sperm DNA isolated from sons of three extensively used US Holstein bulls was screened for differences associated with the primary gene structure of the bovine growth hormone (bGH) and prolactin (bPrl) genes. Southern blot analysis of DNA digested with 10 restriction enzymes revealed that offspring from two of the three bull families exhibited polymorphisms around the bGH and bPrl genes. Restriction fragment length polymorphisms (RFLPs) around the bGH gene were detected with five enzymes, whereas three enzymes … Show more

“…(1987) found RFLPs in the GH gene with the enzymes EcoRI and HindIII. However, our results are in agreement with the findings of Cowan et al (1989) and Hilbert et a/. [ 1989), since these enzymes detect no polymorphisms in this study.…”

“…These RFLPs have been shown to be present in several breeds (Beckmann et ul. 1986;Hallerman et al 1987;Cowan et al 1989;Hilbert et 01.1989;Theilmann et al 1989). In the following the two RFLPs are designated I/D site and MspI site.…”

“…Within the bovine PRL gene, several polymorphisms have been reported (Cowan et al, 1989;Hart et al, 1993;Zhang et al, 1993b;Chung and Kim, 1997 6). The PRL A allele was accepted as the predominant allele in cattle breeds, which is believed to have originated from Bos taurus (Sodhi et al, 2011).…”

“…Within the bovine PRL gene, several polymorphisms have been reported (Cowan et al, 1989;Hart et al, 1993;Zhang et al, 1993b;Chung and Kim, 1997). The genotype and allele frequencies of the PRL gene for the studied cattle breeds are presented in Table 6).…”

“…Leucine or H valine amino acid substitutions at residue 127 in GH exist due to allelic polymorphism (Sartore and Di Stasio 2000). A polymorphic site for Msp 1 restriction endonuclease was detected by Cowan et al (1989) and Hilbert et al (1989) and the polymorphism being localized in intron 3 of the GH gene in position 1,547 (Zhang H et al 1993). Studies on the effect of the GH-Msp1 polymorphism on production traits in Sahiwal cattle are quite advanced, but the results obtained by various workers are not always in agreement.…”
