Resveratrol could reverse the expression of SIRT1 and MMP-1 in vitro

Abstract: ABSTRACT. Intervertebral disc degeneration is the main cause of lumbago disease, in which the extracellular matrix structure and moisture in the nucleus pulposus is lost continuously. In this study, we aimed to detect differential expression of silence mating type information regulation 2 homolog 1 (SIRT1) and matrix metalloproteinase-1 (MMP-1) in human intervertebral disc nucleus pulposus cells and to explore the effects of SIRT1 and MMP-1 on the development of the intervertebral disc degeneration. Interverte… Show more

“…Next, we evaluated if enhancing SIRT1 levels with a pharmacologically relevant compound would prevent skeletal muscle wasting. Previous studies have established that resveratrol (3,5,49-trihydroxystilbene), a natural phytoalexin, increases the deacetylase activity of SIRT1 (Howitz et al, 2003) and increases SIRT1 expression (Dong et al, 2014;Lagouge et al, 2006;Wu et al, 2015). Since we observed that Sirt1 levels were decreased in cachectic muscles, we investigated if resveratrol could revert the cachectic phenotype.…”

“…Sirtuin 1 (SIRT1) is a NAD+-dependent class III histone deacetylase [ 72 ]. The protective effects of SIRT1 against disc degeneration are primarily derived from its abilities to promote ECM anabolism, inhibit inflammatory response and alleviate senescence of CEP cells [ 73 76 ]. It has been demonstrated that SIRT1 markedly decreases the rate of apoptosis in multiple cell types, such as osteoblast-like MC3T3-E1 cells [ 77 ], human kidney proximal tubule epithelial cells [ 78 ], cardiomyocytes [ 79 ], and chondrocytes [ 80 ].…”

“…However, after RSV treatment, the above results were reversed. Therefore, they suggested that RSV promotes ECM expression by altering MMP‐1 and SIRT1 expression, thereby reversing IDD progression 73 . Similarly, Guo et al 72 .…”

“…Previous studies showed that activating Sirtuin 1(SIRT1), a NAD (+)‐dependent deacetylase, could also promote ECM biosynthesis 70–72 . Wu et al 73 . compared the expression of SIRT1 and ECM biosynthesis‐related factors in the NP tissues removed from patients with IDD and lumbar fracture (control group).…”
