Role of gut microbiota and oxidative stress in the progression of non-alcoholic fatty liver disease to hepatocarcinoma: Current and innovative therapeutic approaches

Abstract: Non-alcoholic fatty liver disease (NAFLD) represents the most common chronic liver disease in industrialized countries. NAFLD progresses through the inflammatory phase of non-alcoholic steatohepatitis (NASH) to fibrosis and cirrhosis, with some cases developing liver failure or hepatocellular carcinoma (HCC). Liver biopsy remains the gold standard approach to a definitive diagnosis of NAFLD and the distinction between simple steatosis and NASH. The pathogenesis of NASH is still not clear. Several theories have… Show more

“…Circulating levels of reduced glutathione, a major antioxidant (42,43), was 3-fold higher (P = 3 3 10 24 ) in RS-fed mice compared with NS-fed mice ( Table 1). Plasma glycine, which is necessary for glutathione synthesis, was significantly decreased in RS-fed mice by 0.37-fold (P = 1.6 3 10 22 ).…”

“…Second, we assessed liver oxidative stress by analyzing antioxidant indices and lipid peroxidation biomarkers, including SOD, CAT, GSH-Px, GSH, and MDA. SOD, a critical antioxidant, can convert superoxide radical anions (O 2 − , incompletely reduced forms of oxygen) into hydrogen peroxide (H 2 O 2 ), which in turn is catalyzed into water by CAT and GSH-Px (Turrens, 2003;Borrelli et al, 2018). GSH as a non-enzymatic antioxidant can directly scavenge reactive oxygen species (ROS) by binding with them (Anu et al, 2014).…”

“…Further research is underway to better elucidate the role of gut microbiota on the pathogenesis of NAFLD. 97,98 Journal of Clinical and Translational Hepatology 2019 vol. 7 | 362-370 99 A plethora of studies report an association between NAFLD and these markers of preclinical atherosclerosis, with multiple systematic reviews and meta-analyses highlighting the association between NAFLD and carotid-artery intimamedia-thickness, coronary artery calcification and endothelial dysfunction, independent of traditional risk factors.…”
