Role of protein kinase C-mediated pathway in the pathogenesis of coronary artery spasm in a swine model.

Abstract: These results suggest (1) the PKC-mediated pathway is importantly involved in the pathogenesis of coronary artery spasm, (2) activation of the PKC-mediated pathway partially accounts for serotonin- and histamine-induced coronary artery spasm, and (3) at the spastic site, calcium influx through dihydropyridine-sensitive L-type calcium channel and/or calcium sensitivity of the contractile proteins may be augmented by the PKC-mediated pathway.

“…The cineangiograms were projected on a screen using a cineprojector (ELK-35CB; Nishimoto Sangyo Inc., Osaka, Japan), and an end-diastolic frame was selected and printed. The coronary luminal diameters were measured with a caliper (4)(5)(6)16). Using this technique, excellent correlations between repeated measurements ( r ϭ 0.99) as well as between different observers ( r ϭ 0.98) were confirmed in the range of the coronary diameter from 0.98 mm to 5.58 mm (16).…”

“…A preshaped Judkins catheter was inserted into the right or left femoral artery, and coronary arteriography in a left anterior oblique view was performed. Electrocardiograms (I, II, III, V1, and V6 leads) and blood pressure were continuously recorded throughout the experiments (4)(5)(6)16).…”

“…Ito et al, 11 Katsumata et al, 12 and Kandabashi et al 13 have investigated the intracellular mechanism for spasm using swine coronary spasm models. They indicate that the protein kinase C-mediated pathway and enhanced myosin light chain phosphorylation play an important role in the enhanced constrictor response of the coronary artery smooth muscle to autocoids such as serotonin and histamine.…”
