ROS-dependent HMGB1 secretion upregulates IL-8 in upper airway epithelial cells under hypoxic condition

Abstract: High-mobility group box 1 (HMGB1) mediates various functions according to the location. We tried to investigate the role of HMGB1 in upper airway under hypoxic conditions. We cultured primary normal human nasal epithelium (NHNE) cells under hypoxic conditions and evaluated the movement of HMGB1 by western blotting, immunofluorescence, and enzyme-linked immunosorbent assay (ELISA). Reactive oxygen species (ROS) level was evaluated to estimate the translocation mechanism of HMGB1. The role of secreted HMGB1 was … Show more

“…IL-8 is a neutrophilic chemokine that is freely present in nasal lavage fluid and is a known associate of other DAMP molecules in chronic rhinosinusitis [19,20]. In this experiment (as in other reports), IL-8 was readily detected in all nasal lavage fluids; and although one particular study has demonstrated that in lower airways, baseline CXCL8 mRNA levels of sputum measured in swimmers exceeded those of controls [12].…”

“…2A, 2B). In measuring IL-8 (which was detected and is important in upper airway inflammatory pathways) [20], levels present before (288.94 ± 161.37 pg/mL) and after (319.70 ± 338.49 pg/mL) exercise were similar (Fig. 2C).…”

“…Above results confirmed that ROS and HMGB1 can regulate the p53 and PI3K/Akt pathways. Min et al ( 2017 ) found that hypoxia induced translocation of HMGB1 into the extracellular area and it was dependent on ROS produced by dual oxidase 2. This further proved that ROS can regulate the functions of HMGB1.…”

“…Recently, it has been reported that the alarmin HMGB1 mediates its many activities via an ROS-dependent mechanism. 45 HMGB1 is a known target of such redox reaction molecules, and it contains two redox-sensitive cysteine moieties at positions C23 and C45, the redox states of which greatly affect HMGB1 function. 46 When these cysteine residues are presented in reduced thiol form, HMGB1's prodamage signaling is minimal.…”
