Rutin as A Novel c-Met Inhibitory Lead for The Control of Triple Negative Breast Malignancies

Abstract: Triple negative breast cancer (TNBC) has high metastatic and mortality potential and lacks effective and selective therapeutic options. Aberrant dysregulation of the receptor tyrosine kinase c-Met promotes TNBC progression, motility and survival and therefore considered a valid therapeutic target. Among various identified anticancer agents, plant polyphenols (PPs) including flavonoids, have been shown to be safe and proven for their antitumor activity through modulating diverse macromolecular targets. This stu… Show more

“…Another study has shown that grape stem extract rich in flavonols, particularly rutin, inhibited the growth of MCF-7 cancer cell line [43]. Elsayed et al reported that rutin inhibited c-Met kinase activity in the triple negative breast cancer cell line, significantly reducing the growth of MDA-MB-231/GFP orthotopic xenograft in nude mouse model [44]. Rutin is a common dietary flavonoid found in fruits, vegetables, and plant-derived beverages [45].…”

“…Rutin was also reported to stimulate apoptosis in human breast cancer cells, whereas reinstating the chemosensitivity of HER2 negative and triple-negative breast cancer cells (27). Elsayed et al reported that rutin prevents the activity c-Met kinase in TNBC cells thus considerably reducing the growth of TNBC MDA-MB-231/GFP orthotopic xenografts in murine models (28). Organometallic compounds centered around transition metals have developed as favorable scaffolds in the quest for antitumor leads (29).…”

“…Rutin ( Figure 2 F and Figure 3 ) (rutoside, quercetin-3-O-rutinoside and sophorin), a glycoside combining the flavanol, quercetin, and the disaccharide, rutinose, is widely present in citrus fruits [ 178 , 238 ]. Rutin has anti-carcinogenic, anti-inflammatory, and antioxidant properties [ 178 ].…”
