Recent advances in mammalian cell culture processes have significantly increased product titers, but have also resulted in substantial increases in cell density and cellular debris as well as process and product related impurities. As such, with improvements in titer, corresponding improvements in downstream processing are essential. In this study we have developed an alternative antibody harvest process that incorporates flocculation using a novel stimulus responsive polymer, benzylated poly(allylamine), followed by depth filtration. As tested on multiple antibodies, this process demonstrates high process yield, improved clearance of cells and cell debris, and efficient reduction of aggregates, host cell proteins (HCP) and DNA. A wide operating window was established for this novel flocculation process through design of experiments condition screening and optimization. Residual levels of impurities in the Protein A eluate were achieved that potentially meet requirements of drug substance and thus alleviate the burden for further impurities removal in subsequent chromatography steps. In addition, efficient clearance of residual polymer was demonstrated using a fluorescence tagged polymer in the presence of a stimulus reagent. The mechanism of HCP and aggregates removal during flocculation was also explored. This novel and efficient process can be easily integrated into current mAb purification platforms, and may overcome downstream processing challenges. Biotechnol. This paper reports on an investigation of the effect of initial solid particle concentrationon flock growth and flock shape characterized by the fractal dimension Df2. The experiments werecarried out in a baffled tank agitated by a Rushton turbine at mixing intensity 64W/m3 and constantdimensionless flocculant dosage DF/ck0 = 4.545 mg/g. The model wastewater (a suspension of tapwater and kaolin) was flocculated with Sokoflok 56A organic flocculant (solution 0.1% wt.). The size and shape of the flocks were investigated by image analysis. The flock growth kinetics was fitted according to a semi-empirical generalized correlation proposed by the authors. The dependences Af = 100.35ck01.532, df,eq,max = 1.0474ck0-0.311 and (NtF)max = 1622ck0-0.393 were found. The fractal dimension Df2 was found to be independent of flocculation time and initial kaolin concentration, and its value Df2 = 1.470 ± 0.023 was determined as an average for the given conditions.

Flock growth kinetics was investigated in baffled tank agitated by a Rushton turbine at mixing intensity 40 W/m3 and kaolin concentration 0.44 g/l. The tests were carried out with a model wastewater (a suspension of tap water and kaolin). The model wastewater was flocculated with organic Sokoflok 56 A flocculant (solution 0.1 % wt.). The flock size and flock shape were investigated by image analysis. A simple semiempirical generalized correlation for flock growth kinetics was proposed, and was used for data treatment. The flock shape was characterized by fractal dimension Df2. Using the statistical hypothesis test, the fractal dimension was found to be independent of flocculation time, and the value Df2 = 1.442 ± 0.125 was determined as the average value for the given conditions.
