“…The smooth apposition of the acrosomal membrane to the head is seen. This smooth covering is commonly observed in normal human spermatozoa (Liakatas et al -(1982) but it can also be somewhat wrinkled and disorganized. Furthermore, disorganization of swim-up head membrane was not seen in washed swim-up spermatiozoa.…”

“…Motile swim-up spermatozoa were harvested after two hours from the supernatant and a small fraction of it (0.1 ml) was pelleted in a microcentrifuge tube at 500 x g for 5 minutes. The supernatant was discarded and an electron microscopic fixative (EMF) was layered over sperm pellets, resuspended, fixed overnight at 4 "C and prepared for SEM as previously described (Liakatas et al -(1982). Unwashed semen were fixed similarly and processed for SEM.…”

“…Ultrastructural analysis of sperms by scanning electron microscopy has been proposed by several authors (Gopalkrishnan et al, 1990;Liakatas et al, 1982;Vijayakumar et al, 1987). The disadvantages of this method are small sample volume, limited observation field, as well as complicated and time consuming preparation techniques.…”

“…The disadvantages of this method are small sample volume, limited observation field, as well as complicated and time consuming preparation techniques. However, the benefit this technique offers is that it permits detection of discrete damage to cell surfaces which cannot be achieved with light microscopy or transmission electron microscopy (Liakatas et al, 1982).…”

“…The ability to properly assess morphological sperm features mostly depends on pcrsonal experience and the protocol used in the particular laboratory. Different approaches to determine normal sperm morphology have been described (MacLeod & Gold, 1951;Liakatas et al, 1982). It is difficult to indicate the critical spcrm morphological features that are responsible for fertility potential; therefore the main point of discussion is what a particular feature of sperm morphology may tell a rater in a laboratory or clinic.…”
