Screening and characterization estrogen receptor ligands from Arnebia euchroma (Royle) Johnst. via affinity ultrafiltration LC-MS and molecular docking

Abstract: Arnebiae Radix (dried root of Arnebia euchroma (Royle) Johnst.) is a traditional Chinese medicine (TCM) used to treat macular eruptions, measles, sore throat, carbuncles, burns, skin ulcers, and inflammations. The Arnebiae Radix extract can exert anti-breast cancer effects through various mechanisms of action. This study aimed to rapidly screen potential estrogen receptor (estrogen receptor α and estrogen receptor β) ligands from the Arnebiae Radix extract. In this study, an analytical method based on affinity… Show more

“…This technology utilizes affinity ultrafiltration to separate and enrich active metabolites of TCM that have a strong affinity to specific targets, followed by identification and analysis through mass spectrometry. Zhu et al, 2022 established an analytical method based on affinity ultrafiltration coupled with UHPLC-Q-Exactive Orbitrap mass spectrometry for the rapid screening and identification of active metabolites targeting estrogen receptors (ERα and ERβ). From Arnebiae guttata Bunge ( Boraginaceae ; Arnebiae Radix ), they identified 14 potential active metabolites binding to ERα and 10 potential active metabolites binding to ERβ.…”

“…In particular, ultrahigh-performance LC (UHPLC)–MS has played a pivotal role in detecting constituents of traditional Chinese medicine owing to its heightened sensitivity and selectivity [ 7 , 8 ]. Various studies have used LC–MS to improve detection capabilities for traditional medicinal plant [ 9 , 10 ]. Network pharmacology is recognized for its cost-effectiveness in drug development and is a promising approach for studying compound–proteins/genes–disease pathways, such as the pharmacological mechanisms of traditional Chinese medicine [ 11 ].…”
