Screening Antitumor Bioactive Fraction from Sauromatum giganteum (Engl.) Cusimano & Hett and Sensitive Cell Lines with the Serum Pharmacology Method and Identification by UPLC-TOF-MS

Abstract: Sauromatum giganteum (Engl.) Cusimano & Hett Tuber are used in Chinese folklore medicine for treatment of neoplasms. However, the claim has not been scientifically validated. The aim of the study is to screen the antitumor bioactive fraction of Sauromatum giganteum (Engl.) Cusimano & Hett Tuber and sensitive tumor cell lines using a cytotoxicity assay in vitro and tumor transplantation method in vivo, to support its use in folk medicine. The petroleum ether fraction, chloroform fraction, ethyl acetate fraction… Show more

“…MMP reduction is also involved in oxidative stress, and may result in reactive oxygen species (ROS) release from mitochondria. 27,28 ROS generation in NCI-H460 cells ROS is dynamically equilibrated between antioxidant defense mechanisms and cytotoxic responses leading to cell apoptosis. [29][30][31] In particular, ROS generation could trigger a wide range of transcriptional changes as the second messengers in a signaling cascade, and it might be recovered after leading to apoptosis or cell death.…”

“…Serum pharmacology can objectively simulate the interactions between drugs and the body, and the pharmaceutical ingredients contained in serum are also the endogenous active ingredients produced by the body under the reactions of active ingredients and drugs via a series of in vivo biotransformations. In addition, the physical and chemical properties of serum are similar to that of the internal environments in which the cells are present (14,15). Therefore, if TFA directly acts on cells in vitro, the in vivo metabolic process will remain unclear, thus it cannot truly reflect the pharmacological effects of TFA in vivo.…”

“…Each dry powder (200 g) of TCMs was extracted with 1000 mL cyclohexane (CYH) firstly, and then the leftover solution was sequentially extracted using dichloromethane (DCM), ethyl acetate (EAC), n -butanol (NBA), and distilled water (W), according to the method described by Liu et al [ 13 , 14 ]. The five extracted solutions of 100 TCMs were harvested separately by ultrasonic extraction at 40°C for 30 min and repeated twice followed by filtration and evaporation under vacuum.…”
