Sensitive and specific enzyme-linked immunosorbent assay for the diagnosis of Wuchereria bancrofti infection in urine samples.

Abstract: Abstract. We developed an enzyme-linked immunosorbent assay (ELISA) that detects filaria-specific immunoglobulin G4 antibodies in unconcentrated urine. The ELISA was positive in 87 of 91 (95.6%) urine samples collected from people with Wuchereria bancrofti microfilariae, antigen, or both. Of 298 urine samples collected in Thailand, Lao People's Democratic Republic, and Japan, where no human filariasis is known, 295 (99.0%) were negative by ELISA. Various intestinal nematode and fluke infections did not interfe… Show more

“…In another study, it was shown that the filaria-specific IgG4 titer in urine samples preserved with 0.1% NaN 3 and stored at 4°C did not change within 1 year (unpublished data). Also, filaria-specific IgG4 units in urine samples preserved with 0.1% NaN 3 and stored at 37°C did not change within 4 weeks (21).…”

“…To take advantage of this, several immunodiagnostic methods using urine have been established for some other diseases, like filariasis (21) and schistosomiasis (37). Kohanteb et al (26) reported the detection of soluble antigen and antibody in the urine of VL patients by double-countercurrent immunoelectrophoresis, and de Colmenares et al (11) detected antigenic compounds in urine by the Western blot technique.…”

“…Moreover, several infectious diseases are diagnosed by the finding of specific antibodies in urine, such as Helicobacter pylori infection (18), filariasis (17), or schistosomiasis (35). The presence of anti-Leishmania antibodies in the urine of human patients with visceral leishmaniasis (16,20) and in the urine of L. donovani-infected hamsters (34) has also been described.…”

“…Our study with filarial antibody showed that NaN 3 -added urine samples could be kept at 37 C for 4 weeks without deterioration. 24 The study was reviewed and approved by the Ethics Committee of Aichi Medical University School of Medicine, Japan, and the Ethical Review Committee of the Bangladesh Medical Research Council.…”
