Sequence analysis and transcriptional profiling of two vacuolar H+-pyrophosphatase isoforms in Vitis vinifera

Abstract: Gene expression of grapevine vacuolar H(+)-pyrophosphatase (V-PPase EC 3.6.1.1.) during fruit ripening has previously been reported. Here we report on putative multiple V-PPase isoforms in grapevine. In this study a full-length cDNA sequence with an open reading frame of 2,295 nucleotides encoding a V-PPase gene (vpp2: acc. nr. AJ557256) was cloned. Sequence analyses of the deduced amino acid residues and RT-PCR experiments indicated that Vitis vinifera L. has at least two distinct isoforms of the V-PPase gene… Show more

“…Our RT-qPCR result was consistent with Carystinos's report, which suggested OVP1 increasing at 2 days under chilling stress by western blotting [3]. Expression of vacuolar H þ -translocating pyrophosphatase was also induced by cold stress in grapevine [24] and maize [28]. OVP1 expression pattern indicated that the function of rice V-PPase might be complicated under cold stress.…”

“…3), suggesting its correlation with vigorous growth. In addition, MdVHP1 expressed much higher in mature apple fruits than young fruits, just as reported in grapes (Terrier et al, 1998;Venter et al, 2006). Those findings suggest that MdVHP1 functions in various vegetative and productive tissues, especially in ripened fruit.…”

“…Those findings suggest that MdVHP1 functions in various vegetative and productive tissues, especially in ripened fruit. In addition to the correlation with development and growth, it has been well established in various plants that the expression of VHP genes is induced by abiotic stresses such as high salinity (Fukada et al, 2004;Gao et al, 2006) and cold stress (Venter et al, 2006;Yue et al, 2008;Zhang et al, 2010). Whereas, VHP gene paralogs are differentially regulated in response to abiotic stresses .…”

“…Fruit-specific suppression of the expression of the subunit gene for V-ATPase has previously been shown to affect fruit growth and sugar composition (Amemiya et al, 2005). However, it has also been proposed that VPPase contributes to sugar and organic acid accumulation in fruit cells in grapes, pears, and peaches (Etienne et al, 2002;Suzuki et al, 2000;Venter et al, 2006). Therefore, a detailed expression analysis was performed in tomato fruit prior to demonstrating this role using a transgenic approach (Mohammed et al, 2012).…”
