Sequence analysis of the turkey coronavirus nucleocapsid protein gene and 3′ untranslated region identifies the virus as a close relative of infectious bronchitis virus

Abstract: The 3' end of the turkey coronavirus (TCV) genome (1740 bases) including the nucleocapsid (N) gene and 3' untranslated region (UTR) were sequenced and compared with published sequences of other avian and mammalian coronaviruses. The deduced sequence of the TCV N protein was determined to be 409 amino acids with a molecular mass of approximately 45 kDa. The TCV N protein was identical in size and had greater than 90% amino acid identity with published N protein sequences of infectious bronchitis virus (IBV); le… Show more

“…Both pf/CH/LKQ3/03 and tl/CH/LDT3/ 03 N protein ORFs were 1230 nt in length, within the typical range for most IBV strains and turkey coronaviruses (Boursnell et al, 1987;Breslin et al, 1999b).…”

“…In addition, these analyses demonstrate that the avian coronaviruses, TCoV and IBV, constitute a distinct genotype within the Coronavirus genus. A phylogenetic tree is shown in Figure 1 that compares the complete N protein amino acid sequences of three TCoV strains (Breslin et al, 1999b) with published sequence data for selected IBV strains, and representative group 1 and group 2 mammalian coronaviruses, TGEV and BCV, respectively. This phylogenetic tree demonstrates that TCoV is very closely related to IBV, to such an extent that they cannot be distinguishe d based on this region of the genome.…”

“…They are pleomorphic positive-sense single-stranded RNA viruses measuring 60-220 nm in diameter. 3,11 The characteristic feature of the coronaviral morphology is the presence of long (12-24 nm), widely spaced petal-or pear-shaped surface projections, which impart to the virus the appearance of a solar corona. 14 Three structural proteins have been identified in the virus.…”

“…Infectious bronchitis virus (IBV) and turkey coronavirus (TCoV) belong to group III of Coronaviridae. 3,7,15 Turkey coronavirus causes an acute, highly infec- tious disease affecting turkeys of all ages. The disease is known by various names.…”
