Sequence analysis of three tRNAPhe nuclear genes and a mutated gene, and one gene for tRNAAla from Arabidopsis thaliana

Abstract: Three genes and one mutant gene for tRNA(Phe) (GAA) and one gene for tRNA(Ala) (UGC) were isolated from a whole-cell DNA library of Arabidopsis thaliana. All three tRNA(Phe) genes are identical in their nucleotide sequence, but differ in their 5' and 3' flanking regions. The mutant tRNA(Phe) (GAA) gene differs from the other three genes by one nucleotide change from highly conserved G to C at the 57th nucleotide position. The primary structure of the first tRNA(Ala) gene was also determined in this experiment.

“…Alanine tRNA sequences were amplified by PCR from A. thaliana, rapeseed (Brassica napus) and potato (Solanum tuberosum) nuclear DNA using primers based on the published A. thaliana sequence [ 1]. The 5' primer included the T7 RNA polymerase promoter sequence and the 3' primer included a Bst NI restriction site (CCAGG), such that the final amplification products could be used to produce mature tRNA Ala by in vitro transcription.…”

“…Similarly, tRNA Phe sequences were amplified by PCR from A. thaliana nuclear DNA using primers based on the published A. thaliana sequence [ 1 ] (the major cytosolic tRNA Phe found in most other plant species is identical), cloned into pEMBL18 and sequenced. Again, the primers included the T7 RNA polymerase promoter sequence and a Bst NI restriction site to enable A A in vitro production of functional tRNA transcripts.…”

“…One tRNASer(AGA) gene and two tRNAxyr(GTA) genes occur in tandem arrangement on a 1.5 kb unit which is amplified about 20-fold at a single chromosomal site [6]. Little information exists about the organization of other tRNA gene families in higher plants but apparently different organization patterns are found [1,13,25].…”

“…In a computerassisted similarity search, we found that of plant tRNA species reported to date, tRNAPhe (Arabidopsis) (25) and tRNATrp (wheat) (26) are the most similar to the tRNArelated region of the TS family and they are 64% and 63% identical to this region, respectively (Fig. 3A).…”
