Sequencing and characterization of a gene cluster encoding the enzymes for L-rhamnose metabolism in Escherichia coli

Abstract: The sequencing of the EcoRI-HindIII fragment complementing mutations in the structural genes of the L-rhamnose regulon of Escherichia coli has permitted identification of the open reading frames corresponding to rhaB, rhaA, and rhaD. The deduced amino acid sequences gave a 425-amino-acid polypeptide corresponding to rhamnulose kinase for rhaB, a 400-amino-acid polypeptide corresponding to rhamnose isomerase for rhaA, and a 274-amino-acid polypeptide corresponding to rhamnulose-1-phosphate aldolase for rhaD. “…It would seem that FucA can exhibit a high degree of selectivity in avoiding reaction with 02, but has no greater degree of substrate specificity than RhuA for aldehydic substrates. Although there is some sequence homology between RhuA and FucA (18% identity), they are thought to have arisen by convergent, rather than divergent evolution [22]. Crystals and a crystal structure for both RhuA [23] and FucA [24], respectively, have been obtained.…”

“…The biosynthetic genes involved in the synthesis of dTDP-b-L-Rha in human pathogens such as Salmonella enterica (Marumo et al, 1992), Yersinia enterocolitica (Zhang et al, 1993), Escherichia coli (Moralejo et al, 1993;Marolda and Valvano, 1995), Shigella flexneri (Macpherson et al, 1994), and in plant-associated bacteria including Xanthomonas campestris pv campestris (Koplin et al, 1993) and Rhizobium meliloti (Becker et al, 1997) were discovered and characterized a decade ago. Concomitantly, we identified some Arabidopsis cDNA clones (T44775, R92767, T88368, and Z26952) obtained from the Expressed Sequence Tag (EST) projects at Michigan State University (Newman et al, 1994) and at Institut National de la Recherche Agronomique (INRA), France, that showed sequence similarity to some of these bacterial genes.…”
