“…Until recently, the consensus among most researchers was that Type IIb MyHC, the isoform exhibiting the most rapid speed of contraction, was expressed only in small mammals, in relation to the relative higher speed of contraction of their muscles (Smerdu et al 1994;Tanabe et al 1998;Rivero et al 1999;Pette and Staron 2000). However, the present study reports unequivocally a very high level of expression of the IIb isoform in 100-kg pig L muscle by combining conventional histochemistry, ICC, ISH, and RPA.…”

“…These fibers contain different myosin heavy chain isoforms (MyHCs) that are responsible for their different mATPase activities and speed of contraction. The MyHCs are encoded by a multigene family (Weiss et al 1999;Shrager et al 2000), and four isoforms, i.e., I, IIa, IIx/d, and IIb have been identified in adult skeletal muscle of mice, rats, guinea pigs, and rabbits (Schiaffino and Reggiani 1994;Pette and Staron 2000), whereas only Types I, IIa, and IIx were believed to be expressed in muscles of large mammals (Smerdu et al 1994;Ennion et al 1995;Tanabe et al 1998). However, IIB fibers are metabolically heterogeneous in the pig (Larzul et al 1997) and recent results using in situ hybridization (ISH) suggested a high expression of the IIb isoform in pig glycolytic muscles at 100 kg bw .…”

“…However, an additional fast MHC called "IId" or "IIx" was identified, first in small animals [5,78,94], then in humans [81], pigs [48] and bovines [92]. It was shown to be the product of a specific gene, different from the genes coding IIa and IIb MHC [19,81].…”

“…Recently, Tanabe et al [33] have sequenced 2a, 2x and slow myosin heavy chain isoforms in this species. Therefore, we could speculate that fibres classified as IIB in this species correspond in fact to IIX fibres.…”
