Serotonin is essential for eye regeneration in planaria Schmidtea mediterranea

Abstract: Planaria is an ideal system to study factors involved in regeneration and tissue homeostasis. Little is known about the role of metabolites and small molecules in stem cell maintenance and lineage specification in planarians. Using liquid chromatography and mass spectrometry (LC‐MS)‐based quantitative metabolomics, we determined the relative levels of metabolites in stem cells, progenitors, and differentiated cells of the planarian Schmidtea mediterranea. Tryptophan and its metabolic product serotonin are sign… Show more

“…Serotonin transporters (SERT) terminate signaling by taking up extracellular serotonin. A homolog of the Drosophila peripheral tryptophan hydroxylase (TPH), henna , is highly expressed in both testes and ovaries, relative to the neuronally expressed tph1 (Currie and Pearson, 2013; März et al, 2013; Sarkar et al, 2019). Planarian gonads also express vmat and sert homologs, suggesting the ability to release and recycle serotonin.…”

“…In invertebrates, such as sea urchins, mollusks, starfish, planaria and Drosophila, 5-HT is expressed early in the development and is ascribed various pre-nervous roles [8,11,[79][80][81][82]. In mammals, 5-HT is expressed at different time points in development, for e.g.…”

“…While 5-HT is not known to be supplied via dietary intake or cannot cross the blood-brain barrier, dietary supplementation with tryptophan or 5-hydroxytryptophan an intermediate precursor of 5-HT can raise the blood 5-HT, and since both of these molecules can cross the blood-brain barrier it can increase 5-HT in the brain as well [7]. Moreover, 5-HT is expressed very early in the development in various invertebrates and vertebrates even before the formation of the nervous system and is reported to play a developmental role [8][9][10][11].…”

“…Serotonin inducing contractions of the isolated muscle fibres in planarian Procerodes littoralis was further verified [63]. Serotonin also stimulated the development of metacestodes of E. multilocularis [64], regulating RNA synthesis in flatworms [65] and was essential for eye regeneration in planarians S. mediterranea [66] and Girardia tigrina [67], as well as accelerated growth of planarian regenerative blastema in [68]. (Kreshchenko, personal report).…”
