Serotonin–mushroom body circuit modulating the formation of anesthesia-resistant memory in Drosophila

Abstract: Pavlovian olfactory learning in Drosophila produces two genetically distinct forms of intermediate-term memories: anesthesia-sensitive memory, which requires the amnesiac gene, and anesthesiaresistant memory (ARM), which requires the radish gene. Here, we report that ARM is specifically enhanced or inhibited in flies with elevated or reduced serotonin (5HT) levels, respectively. The requirement for 5HT was additive with the memory defect of the amnesiac mutation but was occluded by the radish mutation. This re… Show more

“…A wealth of evidence suggests that aversive olfactory memory exists as a persistent neural activity ("trace") in a sparse subset of MB neurons-initially in the γ lobes lasting for several minutes and then spreading to α/β lobes for several hours (34)(35)(36). This physiological trace drives a protein-synthesis-dependent process in postsynaptic MB-V3 neurons, yielding structural change at MB::MB-V3 synapses (Fig.…”

“…Cantonized w 1118 [w(CS10)] or Canton-S flies of mixed sex were used as normal controls in our experiments. Flies carrying uas transgenes included uas-shi ts (Kitamoto, 2001) anduas-G-CaMP3.0 (Tian et al, 2009). Gal4 driver lines included c316-gal4, c739-gal4, and VT64246-gal4.…”

“…Functionally, the different MB neurons seem to participate in different aspects of memory processing, as revealed through behavioral studies, although this relationship is complex and not fully understood. Both the ␥ and ␣Ј/␤Ј neurons appear to have roles in early forms of memory, whereas the ␣/␤ MB neurons are involved in longer forms of memory and retrieval (Yu et al, 2006;Krashes et al, 2007;Wang et al, 2008;Akalal et al, 2011;Qin et al, 2012;Cervantes-Sandoval et al, 2013). Furthermore, memory traces with different temporal features, as revealed by functional cellular imaging, have been mapped to different classes of MB neurons (Yu et al, 2006;Wang et al, 2008;Akalal et al, 2010;.…”
