Serotype 6B from a pneumococcal polysaccharide vaccine induces cross-functional antibody responses in adults to serotypes 6A, 6C, and 6D

Abstract: Cross-reactivity of pneumococcal capsular polysaccharides is a key element for formulating pneumococcal vaccines and evaluating vaccine efficacy. This study examined whether 23-valent pneumococcal polysaccharide vaccine (PPSV23), which only contains 6B, can elicit cross-functional immune responses against recently discovered serotypes (6C and 6D), as well as against 6A, in 2 adult age groups.Young adults (25–51 years; N = 28) and elderly subjects (over 65 years; N = 60) were immunized with PPSV23. Functional a… Show more

“…In another study of elderly adults, post-vaccination IgG antibody concentrations for two serotypes (6B and 19F) of the five studied (4, 6B, 14, 19F, and 23F) were significantly lower and in vitro opsonophagocytosis of all five IgG serotype-specific antibodies was significantly reduced [50]. In two other reports, serotype-specific pre- and post-immune antibody responses measured by opsonophagocytosis were also significantly lower in elderly adults [51, 52]. IgG responses to PPS4 and PPS14 in elderly adults were associated with decreased V H repertoire (mostly V H 3) and somatic mutations and with greater loss of oligoclonality than younger adults [51].…”

“…The serotype-specific VE was estimated for each serotype if its number of isolates was >30. To explore the potential effect of cross-immunity produced by PPSV23 on serogroup 6 ( 23 ), VE of IPD for serotypes 6A, 6B, 6C, and 6D was calculated excluding these serotypes from the controls. VE was stratified by the 2 study phases (2013–2015 and 2016–2017).…”

“…Furthermore, 35 sera from healthy adults (30–50 years old) and 62 sera from the elderly (65–85 years old) were studied after receiving each subject's written informed consent. 12 In the elderly, subjects with pre-existing but stable health conditions (including well controlled diabetes mellitus, cardiovascular disease, pulmonary disease, and liver cirrhosis) were included. However, subjects with immune-compromising conditions such as human immunodeficiency virus (HIV) infection, leukemia, generalized malignancy, chronic renal failure, primary or acquired immunodeficiency, and diseases requiring immunosuppressive-drug treatments were excluded.…”

“…However, subjects with immune-compromising conditions such as human immunodeficiency virus (HIV) infection, leukemia, generalized malignancy, chronic renal failure, primary or acquired immunodeficiency, and diseases requiring immunosuppressive-drug treatments were excluded. 12 Collected samples were stored frozen at −70°C until testing. All sera were found to be free from antibiotics based on their inability to inhibit the growth of R36A pneumococcal bacteria, and were incubated at 56°C for 30 minutes to inactivate endogenous complement activity before our opsonophagocytic killing assay (OPA).…”
