Toxicodynamic Effects of ‘Red Bull’ Energy Drink in A Randomised Controlled Study on Local Strains of Adult Rabbits

Increasing reports of anecdotal cases of acute adverse effects associated with the consumption of energy drink have been expressed by international bodies. This has necessitated the need to assess toxicological effects of Red Bull (RB) energy drink in this study. Eighteen adult rabbits were randomly divided into 3 groups of 6 rabbits (3 males and 3 females). Rabbits in groups 1, 2 and 3 (control) received daily oral doses of 23.1 ml RB, 11.5 ml RB and water respectively for 21 days. Venous samples were collected on days 1 (baseline), 14, 21 and 28, and animals were sacrificed on days 21 and 28. Biochemical assessment of liver and renal functions, and muscle activity were carried out by spectrophotometry, in addition to haematological and histological studies. Mortality of 11.1% was recorded, involving 2 female rabbits in group 1. For male rabbits in this group, significant increase (p<0.05) in total protein (TP) concentration and in activity of ALT and ALP occurred on days 14 and 21, with significant decrease in platelet counts. For female rabbits in group 2, significant increases in TP concentration and ALT activity were observed with significant decrease in albumin concentration and AST activity. Apart from slight increase in ALP activity of female rabbits, there were no significant differences in mean values of all rabbit parameters measured in the control group. Histology of rabbits in group 1 showed congestion and balloon degeneration of hepatocytes. This result showed that administration of RB to rabbits at doses lower than the manufacturer's recommended equivalent daily human intake of 500 ml caused adverse effects. There is need for caution in consumption of this drink.

This study aimed to address the different gamma radiations doses effect on the liver and kidney function of rats: in vivo. A total of 60 healthy male Wistar-Kyoto rats were whole body gamma irradiated with Co 60 source with 0.883 cG/sec dose rate at the beginning of the experiment. The rats were randomly divided into 4 gamma-irradiation groups (25, 50, 75 and 100 Gy) The serum levels of activity of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyltransferase (GGT), UREA, creatinine (CREA) and uric acid (UAC) were measured using automated biochemical analyzer. The ALT, GGT, ALP values significantly decreased with the different gamma radiation doses compared with the control. The AST and UREA values significantly decreased after irradiation with 25 and 50 Gy gamma radiation doses compared with the control while it significantly increased with 75 and 100 Gy gamma radiation doses. The levels of CREA values decreased with no significant manner after the irradiation with gamma radiation doses compared with the control. The levels of UAC values significantly increased with 50, 75 and 100 Gy gamma radiation doses. The serum ALT and AST levels are common markers for hepatic toxicity: A lower amount of ALP indicates liver problems. The decreased CREA and the increased UAC levels might indicate development of nephritis and renal dysfunction. The excess UAC might be converted to crystals depositing in the tiny tubes of the kidney and causing acute kidney damage. It is proposed that oxidative stress is linked to the organ damage following exposure to ionizing radiation, and after the onset of oxidative stress, antioxidant treatment should be applied to delay or prevent the progression of damage.

Context: Purslane (Portulaca oleracea L., Portulacaceae) has been traditionally used in folk medicine to afford protection against liver injury, although its actual efficacy remains uncertain. Objective: To evaluate purslane as a hepatoprotective agent, we investigated the protective effect of its ethanol extract against carbon tetrachloride (CCl 4 )-induced hepatic toxicity in rats. Materials and methods: A total of 108 male Wistar rats were randomly divided into 12 groups. The first group was maintained as normal control, whereas CCl 4 (0.5 ml/kg bw, 50% CCl 4 in olive oil, i.p.), purslane extract (0.005, 0.01, 0.05, 0.1, and 0.15 g/kg bw, intragastrically), and purslane extract (five doses as above) along with CCl 4 were administered to the Groups II, III-VII, and VIII-XII, respectively. The rats were sacrificed on the 30th day, and blood was withdrawn by cardiac puncture. Liver damage was assessed by measuring hepatic marker enzymes (ALT, AST, ALP, GGT, and SOD) and histopathological observation. Results: Treatment with CCl 4 resulted in increased serum activities of marker enzymes with a concomitant decrease in SOD. Histological alterations were also observed in the liver tissue upon CCl 4 treatment. Administration of purslane extract (0.01, 0.05, 0.1, and 0.15 g/kg b.w.) significantly showed a marked tendency towards normalization of all measured biochemical parameters in CCl 4 -treated rats. Histopathological changes also paralleled the detected alteration in markers of liver function. Discussion and conclusion: These results demonstrate that purslane exerts protective effects against CCl 4 -induced damage in rat liver and supports a potential therapeutic use of purslane as an alternative for patients with liver diseases.
