SETDB1 plays an essential role in maintenance of gonocyte survival in pigs

Abstract: Histone methyltransferase SETDB1 suppresses gene expression and modulates heterochromatin formation through H3K9me2/3. Previous studies have revealed that SETDB1 catalyzes lysine 9 of histone H3 tri-methylation and plays essential roles in maintaining the survival of embryonic stem cells and spermatogonial stem cells in mice. However, the function of in porcine male germ cells remains unclear. The aim of the present study was to reveal the expression profile and function of in porcine germ cells. expression gr… Show more

“…SIRT1 may regulate granulosa cell apoptosis during follicular atresia in porcine ovaries [39] and can reduce ovarian cell viability in rats [40]. SETDB1 plays an essential role in the maintenance of gonocyte survival in pigs, and knockdown of SETDB1 can induce gonocyte apoptosis [41]. PSMD4 effectively inhibits sperm-oocyte binding [42].…”

“…To investigate the function of SETDB1 in porcine SSCs, we knocked down SETDB1 in porcine SSCs by siRNAs [31,37]. The immortalized porcine SSC line was established by stably expressing the simian virus 40 (SV40) large T antigen (Figure 2A) [31].…”
