Sialic Acids in the Immune Response during Sepsis

Abstract: Sialic acid-binding immunoglobulin-type lectins (Siglecs) are a group of cell surface transmembrane receptors expressed on immune cells, and regulate immune balance in inflammatory diseases. Sepsis is a life-threatened inflammatory syndrome induced by infection, and the pathogenesis of sepsis includes immune dysregulation, inflammation, and coagulation disorder. Here, we reviewed the various roles acted by Siglecs family in the pathogenesis of sepsis. Siglec-1, Siglec-5, and Siglec-14 play bidirectional roles … Show more

“…Hence, many lines of evidence indicate that Gal3‐dependent interactions may allow metabolic flux through the HBP pathway to control both cell surface as well as intracellular membrane traffic. Notably, the regulation of receptor traffic by other lectin‐dependent interactions is far broader and an area of research with much promise to reveal more intricate levels for control of endomembrane traffic, such as that resulting from control of N‐ and O‐glycans and their lectin‐glycan interactions by cell metabolic determinants. In cancer cells, the availability of receptor cargo for interaction with Gal3 may increase, as Mgat5 is upregulated in certain cancers and many cancer cells exhibit increased flux through HBP and elevated UDP‐GlcNAc levels .…”

“…The interaction between Siglec-10 expressed on human DC and pseudaminic acid can increase the expression of IL-10 through MyD88 and p38 MAPK signaling pathway to promote anti-in ammatory function [19]. Several other reports also con rmed its ability to in uence the in ammatory microenvironment [20][21][22][23]. Research on tumors indicates that tumors achieve immune escape through the inhibition of NK cells and T cells and reduction of the tumor-killing ability of macrophages, which comes out from up-regulating the expression of Siglec-10 [24,25].…”

“…Sialic acid-binding immunoglobulin-like lectins (Siglecs) are a group of cell-surface transmembrane receptors expressed on immune cells that regulate the immune equilibrium in inflammatory diseases 159 . Homozygotes for the null allele in SIGLEC14 are associated with a reduced risk of exacerbation in a population of Japanese patients 160 , while allele G of rs2075803 and haplotype GA (rs2075803 G/rs2258983 A) in SIGLEC9 were associated with a higher frequency of exacerbations and with the emphysema level 161 .…”
