Side Effects on the Skeletal System Exerted by Continuous and by Interval Treatment with Tyrosine Kinase Inhibitors in An Animal Model of Juvenile Rodents,

Abstract: 3741 Background: Chronic myeloid leukemia (CML) is a rare malignancy in children. The oncogenic transformation of hematopoietic stem cells results from the dysregulated tyrosine kinase (TK) activity of the BCR-ABL fusion protein. As front line treatment the TK inhibitor (TKI) imatinib (IMA; Gleevec®, Novartis) has radically changed the hematological, cytogenetic and molecular response rates in adults as well as in children with CML. IMA blocks the BCR-ABL onc… Show more

“…In a juvenile animal model, male 4 week-old Wistar rats were exposed to varying concentrations of the 3 TKIs imatinib, dasatinib and bosutinib, respectively, via the drinking water chronically over 10 weeks. Rats from this strain enter weaning when 3 weeks old, puberty when 7-8 weeks old, and start their reproductive phase at an age of 9-10 weeks [ 36 ] . Rats were purchased from a commercial supplier (Elevage Janvier; Le Genest St. Isle, France) and kept in cohorts under standardized conditions and 12 h lightning (7:00-19:00 hrs) with free access to food (V1534-000, Ssniff Spezialdiaeten, Soest, Germany) and water.…”
