Simian virus 40 large tumor antigen is unable to transform mouse embryonic fibroblasts lacking type 1 insulin-like growth factor receptor.

Abstract: Fibroblast cell lines were established from mouse embryos homozygous for a targeted disruption of the lgflr gene, encoding the type 1 receptor for insulin-like growth factor I (IGF-I) and from their wild-type littermates. The cells from the wild-type embryos (W cells) grow in serum-free medium supplemented with platelet-derived growth factor, epidermal growth factor, and IGF-I, whereas the cells from Igflr(-/-) embryos (R-cells) do not, although they grow at a reduced rate in 10% fetal calf serum. The simian v… Show more

“…The ®nding that the complete absence of the IGF-IR inhibits transformation (Sell et al, 1993) is not invalidated by the present results, since a dominant negative mutant of the IGF-IR still causes apoptosis of LNCaP cells . The picture that emerges is that the IGF-IR (even at low levels) is crucial for transformation, but that it requires IRS-1 for this function.…”

“…In transformed MEFs with high IGF-IR numbers, like R þ cells (Sell et al, 1993), IRS-1 is largely nuclear (Tu et al, 2002). In cells with lower numbers of IGF-IR (R12 cells, and other cell types), IRS-1 is confined to the cytoplasm.…”

“…By ChIPs, IRS-1 is detectable in the rDNA promoter (Chen et al, 2005). IGF-1 fails to translocate IRS-1 to nuclei of RÀ cells (Tu et al, 2002) that are MEFs with a targeted disruption of the IGF-IR genes (Sell et al, 1993). We asked whether nuclear IRS-1 in R þ cells would also increase transcription from RNA polymerase II promoters.…”
