Simple, Rapid and Sensitive UV-Visible Spectrophotometric Method for Determination of Antidepressant Amitriptyline in Pharmaceutical Dosage Forms

Abstract: The paper describes a new and simple approach for spectrophotometric determination of tricyclic antidepressant drug amitriptyline. Enhancement of the colour intensity of the Fe(III)-SCN−complex on addition of the drug amitriptyline forms the basis of the proposed method. The value of molar absorptivity of the Fe(III)-SCN−amitriptyline ion pair complex in terms of the drug lies in the range of (2.82–3.36) × 103 L·mol−1·cm−1at the absorption maximum 460 nm. The detection limit of the method was 0.3 μg·mL−1. The … Show more

“…Citalopram hydrobromide is marketed as Cipramil, and its active ingredient is citalopram [ 13 , 14 , 15 ], which is a selective serotonin reuptake inhibitor. Some studies have shown that the oral medication compliance of patients with depression is low, at only 40–70% [ 16 ], and oral medication cannot achieve good treatment effects. The oral administration of citalopram hydrobromide (CTH), a commonly used antidepressant, can cause gastrointestinal irritation, such as nausea, vomiting, diarrhea, constipation, appetite change, and dry mouth, affecting patient compliance and quality of life [ 17 ].…”

“…The present study also describes the effective use of this method for assessing the concentration of the active ingredient in various pharmaceutical formulations and preparations available on the European market, as well as comparisons with other similar spectrophotometric methods. Several analytical techniques have been developed to quantify TCAs, including highperformance liquid chromatography [18][19][20][21][22], gas-liquid chromatography [2,23], cyclic voltammetry [24], capillary electrophoresis [25,26], and ultraviolet-visible (UV-VIS) spectrophotometry [27][28][29]. UV-VIS spectrophotometry is widely regarded as a favorable method for quantifying various chemical compounds due to its many benefits such as its simplicity, cost-effectiveness, little reagent usage, and adaptability [30][31][32][33].…”

“…AMT can be determined via analytical techniques such as gas chromatography (GC), 5 liquid chromatography (LC), 6 high performance liquid chromatography (HPLC), 7 capillary electrophoresis (CE) 8 and UVvisible spectrophotometry. 9 These techniques are typically only used in the laboratory, where they provide very reliable and acceptable results. However, these methods are not without limitations: instrument costs are high (especially for chromatographic techniques), expert analysis is required, and the process is time-consuming.…”
