Simple strategy for the in vitro conservation of Alnus glutinosa (L.) Gaertn. germplasm

Abstract: The aim of this study was to develop a simple method for the medium-term storage of Alnus glutinosa (L.) Gaertn. explants obtained from trees aged 20-30 years. Several parameters were evaluated, including type of explant (shoot apex or nodal segments), pre-storage treatment (0 or 10 days after the last subculture) and duration of cold storage (3-24 months) at 2-4ºC. Explants were maintained at this temperature under dim lighting on Woody Plant Medium supplemented with 0.1 mg l-1 6-benzyladenine and 0.5 mg l-1 … Show more

“…Por tanto la aplicación de estas herramientas biotecnológicas con fines de conservación se justifican para proteger la diversidad genética de las especies (Mohanty et al 2013;San José et al 2014). En este trabajo se evaluará la efectividad de la modificación del medio de cultivo y la adición de agentes osmótico en la conservación in vitro para limitar el crecimiento y del nitrógeno líquido (NL) (-196°C) en la conservación a largo plazo de explantos de C.…”

“…On the other hand, tissue cryopreservation involves the suspension of growth for indefinite periods (Paunescu 2009;Ai et al 2012;Reed et al 2011;Sen-Rong and Ming-Hua 2012;Cruz et al 2013;Panis et al 2020;Popova et al 2023). The application of these techniques translates into a significant contribution to the conservation of species facing challenges in natural propagation (Ruta et al 2020), safeguarding the genetic diversity of the species in question (Mohanty et al 2013;San José et al 2014;Armijos-González et al 2021).…”

“…The in vitro technique is a powerful tool to maintain plant genetic resources. It has been widely used for a long time as an efficient alternative to preserve native and cultivated plant germplasm [1,2] and also to quickly increase the number of plants of commercial interest [3]. In addition, the possibility to modulate the production of chemical components constitutes an interesting aspect to explore in vitro cultivation [4,5,6].…”
