Simultaneous detection of assembly and disassembly of multivalent HA tag and anti‐HA antibody in single in‐capillary assay

Abstract: Herein, we have developed an in-capillary assay for simultaneous detection of the assembly and disassembly of the multivalent HA tag peptide and antibody. HA tag with hexahistidine at C terminus (YPYDVPDYAG4 H6 , termed YPYDH6 ) was conjugated with quantum dots (QDs) by metal-affinity force to form a multivalent HA tag (QD-YPYDH6 ). QD-YPYDH6 and monoclonal anti-HA antibody (anti-HA) were sequentially injected into the capillary. They were mixed and assembled inside the capillary. The reaction products were on… Show more

“…The same instrumentation, methodology, and BGE were employed for investigation of a large series of interactions (self‐assembly) between oligo‐ and polypeptides of different lengths and charges (mostly tagged with hexahistidine anchor) and CdSe/ZnS quantum dots or between peptides conjugated to these quantum dots and other peptides or proteins, e.g. characterization of self assembly of glutathione stabilized CdSe/ZnS quantum dots and cyanine5‐labeled peptide (cyanine5‐Glu(Ala) 5 (His) 6 ) , detection of assembly and imidazole‐induced disassembly of multivalent HA tag peptide with hexahistidine at C‐terminus and anti‐HA monoclonal antibody , and study of multivalent interactions between conjugate of quantum dots with c‐Myc peptide tag and the anti‐c‐Myc monoclonal antibody . In the last two cases, the assays provided also the dissociation constants and stoichiometry of the complexes.…”

“…Expression of eGFP did not prevent cell division and was persistent in cell division, like HEV [9]. Since its introduction almost 20 years ago as a short immunoreactive tag, the hemagglutinin epitope YPYDVPDYA (HA tag) from influenza virus A has been used to track proteins of interest within cells, to isolate them, and to coprecipitate their possible partners [10,11]. Using the HA-tag replicons, HA-tag protein cloned into aHEV was expressed in LMH cells.…”
