Simultaneous detection of severe acute respiratory syndrome, Middle East respiratory syndrome, and related bat coronaviruses by real-time reverse transcription PCR

Abstract: Since severe acute respiratory syndrome (SARS) and Middle East respiratory syndrome (MERS) coronaviruses (CoVs) share similar characteristics with respect to clinical signs, etiology, and transmission, methods for a rapid and accurate differential diagnosis are important. Therefore, the aim of this study was to develop a duplex real-time reverse transcription (RT)-PCR method for the simultaneous detection of these viruses. Primers and probes that target the conserved spike S2 region of human SARS-CoV, MERS-CoV… Show more

“…Real-time reverse transcriptase-PCR (RT-PCR) detection is currently favored for the detection of coronavirus because of its advantages as a specific, and simple quantitative assay. Moreover, realtime RT-PCR is more sensitive than the conventional RT-PCR assay, which help much for the diagnosis in early infection [10,11]. Therefore, the real-time RT-PCR assay still is a predominant method to be applied for the detection of all kinds of coronaviruses [12,13], including SARS-CoV-2 [14].…”

“…A duplex RT‐PCR method has been developed based on primers and probes targeting the conserved spike S2 gene of SARS‐CoV and MERS‐CoV. By using pUC57SARS‐pS2 as a template for SARS‐CoV and pGEM‐MERSS2 as a template for MERS‐CoV, respectively, adequate detection limits of 50 to 100 copies/mL were achieved 28 . The singleplex RT‐iiPCR assays are designed to detect MERS‐CoV envelope gene (upE) and open reading frame 1a (ORF1a) genes separately.…”

“…Reverse transcription polymerase chain reaction (RT-PCR) is a technology combining RNA reverse transcription (RT) with polymerase chain amplification (PCR) of cDNA. A duplex RT-PCR assay can be used to detect SARS-CoV and MERS-CoV using pUC57SARS-pS2 and pGEM-MERSS2 as templates, respectively [12]. Also, samples collected from the upper respiratory tract (oropharyngeal and nasopharyngeal) and lower respiratory tract (endotracheal aspirate, expectorated sputum, or bronchoalveolar lavage) of suspected 2019-nCoV patients can be diagnosed by RT-PCR [8].…”
