Single-Cell Transcriptome Analysis Reveals the Role of Pancreatic Secretome in COVID-19 Associated Multi-organ Dysfunctions

Abstract: The SARS-CoV-2 infection affects the lungs, heart, kidney, intestine, olfactory epithelia, liver, and pancreas and brings forward multi-organ dysfunctions (MODs). However, mechanistic details of SARS-CoV-2-induced MODs are unclear. Here, we have investigated the role of pancreatic secretory proteins to mechanistically link COVID-19 with MODs using single-cell transcriptome analysis. Secretory proteins were identified using the Human Protein Atlas. Gene ontology, pathway, and disease enrichment analyses were us… Show more

“…Because of the high replication rate, a SARS-CoV-2 infection generates more than one million virions in one day [ 80 ], and most known antivirals are ineffective against it due to SARS- CoV-2's inherent genetic mutability [ [84] , [85] , [86] ]. COVID-19 patients showed respiratory symptoms, multi-organ dysfunction, and inflammation [ [2] , [3] , [4] ]. Therefore, in comparison to single target approaches, concurrent inhibition of multiple SARS-CoV-2 targets [ 87 , 88 ] combined with inflammation and immunomodulation associated with tissue-specific host targets may be more effective and have a significantly higher therapeutic potential in saving the life of a COVID-19 patient.…”

“…There had been 537,591,764 confirmed instances of COVID-19 documented as of June 22nd, 2022, resulting in 6,319,395 deaths globally [ 1 ]. COVID-19 patients exhibit respiratory symptoms, multi-organ failure, and inflammation, and therapeutic options are currently limited [ [2] , [3] , [4] ]. COVID-19 continues to spread over the world despite the availability of vaccines to the wider population, fueling the need for effective therapeutics to prevent and treat COVID-19 [ 5 ].…”

“…Their livers showed the enrichment of processes involved in the acute phase response, cytokine secretion and neutrophil degranulation. To gain a better understanding of the mechanisms exerted by SARS-CoV-2, the authors selected six functional protein clusters, including viral receptors and proteases, transcription factors (TFs), cytokines and their receptors, the complement and coagulation system, angiogenesis and fibrosis markers, as reported in other studies [ 29 , 30 , 42 , 45 , 46 ]. TFs were strongly correlated with the activation of the inflammatory response, and 395 out of 1117 were found altered; they were enriched also in processes involved in the spliceosome, viral carcinogenesis, tissue injury and hypoxia.…”

“…In the patient's pancreas samples, scientists observed the overexpression of 102 genes responsible for damage to the pancreas and myeloid leukocyte migration pathways. Moreover, an imbalance of pancreatic secretory proteins was associated with dysfunction of the RAS pathway, which resulted in overexpression of cytokines including TNF-α, interleukins -1 and -6, and CAM [138]. Taking into consideration the increased level of pro-inflammatory cytokines, Qu et al published data from two COVID-19 patients treated with tocilizumab.…”
